Abstract: Based on the activity of amino contained in chemical products, it can participate in a variety of reactions, such as reaction with acid to form salt, acylation of amine (acyl halide, acid anhydride, carboxylic acid, ester can react with ammonia to obtain amide) and aldehyde-ketone reaction, etc., in order to meet the requirements of the target product, it is usually necessary to protect the amino from participating in the reaction. Several common methods of amino protection and deprotection were comprehensively introduced, and the reaction conditions and yield of each method were listed; five kinds of methods, including acylation, alkylation, alkoxy carbonylation, protonation reaction and chelation reaction, and their related examples were summarized, and their advantages and disadvantages were evaluated. The research is helpful to improve the application protection and deprotection of amino in chemical products, and provides a reference for related compounds from exploring synthesis conditions in the laboratory to factory production.
