Determination of the Content of Fusidic acid in Fermentation Broth by HPLC Method

HPLC( High performance liquid chromatography ) method to determine the content of the fusidic acid in fermentation broth was developed. The HPLC was carried out with Agilent Extend-C18 column. The mobile phase was phosphate buffer -acetonitrile ( 30∶70 ) with a flow rate of 1.2 mL·min-1. The detection wavelength was set at 214 nm and the column temperature was 30 ℃. The results show that the concentration of fusidic acid ranges in 10 -2 000 μg·mL-1 ,the linear relationship between peak area and mass concentration is good, the correlation coefficient is 0.9991. According to standard addition method, the average recovery is 99.56% and the precision is 1.13%. The method is rapid in analysis, convenient in operation, accurate in result and good in reproducibility etc., and is suitable for a large number of strain screening and the online monitoring of refinery and purification.
