A high-throughput screening method for common Ketogulonicigenium vulgare was established to achieve rapid detection strains with fast-growing and high-yield of 2-keto-L-gulonic acid (2-KGA) . Mixed indicators were added to liquid and solid media respectively, and the strains with relatively high acid production were screened according to the color change; mass culture and detection were carried out by centrifugal tube group culture technology. The experimental results show that the discoloration condition, the size of the discoloration circle and the discoloration degree of the liquid medium around the single colony of the medium around a single colony on a solid plat were a certain corresponding relationship with the metabolic acidity of the common keto producing gulonic acid bacteria. The strains with fast growth and high acid production could be directly detected by visual inspection, and the 2-KGA high-yield strain K56-1 was screened. Compared with the control strain, the fermentation period of K56-1 was shortened by about 2.38 h, and the production rate of 2-KGA was increased by 7.23%.
