Separation and purification of melittin from bee venom

Abstract:The purification process of melittin from bee venom was developed to obtain high-purity melittin. The crude bee venom was pretreated by centrifugation and ultrafiltration, combined with preparative liquid chromatography technology, polystyrene microspheres were used as fillers, the type of filler, mobile phase and sample loading were investigated, combined with preparative liquid chromatography thchnology. The experimental results showed that the separation and purification effect of melittin was better when the sample loading amount was 1.2% with polymer microspheres PS20-300 as filler and acetonitrile/water containing 0.1% trifluoroacetic acid as the mobile phase for elution. The optimized chromatographic conditions were amplified and prepared, the purity of melittin was 99.2% and the purification yield was 58.9%.
