Abstract

Insulin-like growth factor-1(IGF-1) is involved in cell proliferation, differentiation and apoptosis in a wide range,and has also been shown to be associated with some types of normal and diseased physiological processes. The recombinant human IGF-1 has been used to treat children dwarfism caused by severe deficiency of primary IGF-1. In order to expand range of applications and safety and effectiveness of IGF-1, researchers are actively developing IGF-1 derivatives and analogs. IGF-1 derivatives developed through a number of technological strategies were introduced which included truncated IGF-1 and lengthened IGF-1, or the fusion of the Fc domain of IgG1 to IGF-1 and IGF-1 derivatives, or construction of pegylated IGF-1. It is pointed out that based on the modifiable molecular structure and pharmacokinetic and pharmacodynamic properties of recombinant IGF-1 derivatives, it is hopeful that more IGF-1 drugs with prolonged half-life and increased bioactivity will be developed to meet the modern clinical demands.
